应用免疫组化和同位素点杂交技术对 2 5例甲状腺乳头状癌新鲜冰冻组织及 19例其他类型甲状腺肿瘤和非肿瘤组织中 Neu癌基因 p185蛋白和 m RNA的表达进行研究。乳头状癌中 17例 p185癌蛋白表达阳性 ,而其他类型肿瘤和非肿瘤组织均未见 p185表达 ;乳头状癌和淋巴结转移灶中 Neu癌基因 m RNA的表达水平明显高于其他肿瘤和非肿瘤组织。而且石蜡包埋的乳头状癌标本中 Neu癌基因蛋白表达率明显低于新鲜标本。认为 Neu癌基因 p185蛋白的表达是甲状腺乳头状癌的一个特征性改变 ,Neu蛋白的表达与 m RNA的表达相一致 ,Neu癌蛋白检测的稳定性取决于组织标本的质量。
The expression of p185 protein and m RNA in 25 cases of fresh frozen tissue of thyroid papillary carcinoma and 19 cases of other thyroid tumors and non-tumorous tissues were studied by immunohistochemistry and dot blot hybridization. The positive expression of p185 oncoprotein was positive in 17 cases of papillary carcinoma, while the expression of p185 was not found in other types of tumors and non-tumorous tissues. The expression of m RNA in tumor tissues and lymph node metastases was significantly higher than that of other tumors organization. Moreover, the expression of Neu oncogene protein in paraffin-embedded papillary carcinoma specimens was significantly lower than that of fresh specimens. It is considered that the expression of the p185 protein of Neu gene is a characteristic change of thyroid papillary carcinoma. The expression of Neu protein is consistent with the expression of m RNA. The stability of detection of Neu oncoprotein depends on the quality of tissue samples.
