目的检测骨桥蛋白(OPN)mRNA在卵巢组织中的表达及其在卵巢癌转移发生、发展中的意义。方法采用半定量反转录-聚合酶链反应(RT-PCR)法检测46例卵巢病变组织,30例相应癌旁卵巢组织、18例卵巢癌转移淋巴结、30例卵巢良性病变及79例正常卵巢组织中OPNmRNA的表达,并评价其与临床病理特征的关系。结果 OPNmRNA表达用OPNmRNA量与β-肌动蛋白mRNA量的比值来表示:OPNmRNA在卵巢癌组织及转移淋巴结组织中表达最高,分别为1.23±0.13,1.15±0.09;良性病变卵巢组织次之,比值为1.09±0.11;而在癌旁卵巢组织及正常卵巢组织中表达最低,比值为0.32±0.07,0.21±0.04,差异有统计学意义(P<0.05)。结论 OPN的表达与卵巢癌恶性程度有关,并可能与卵巢癌的发生有关,检测其表达水平,可望能指导临床治疗,评判患者预后。
Objective To detect the expression of osteopontin (OPN) mRNA in ovarian tissue and its significance in the occurrence and development of ovarian cancer. Methods 46 cases of ovarian lesions, 30 cases of corresponding para-ovarian tissue, 18 cases of ovarian cancer metastatic lymph nodes, 30 cases of benign ovarian lesions and 79 cases of normal ovarian tissue were detected by semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) The expression of OPNmRNA in tissues and its relationship with clinicopathological features were evaluated. Results The expression of OPN mRNA was expressed as the ratio of OPN mRNA to β-actin mRNA. The expression of OPN mRNA was the highest in ovarian cancer tissues and metastatic lymph nodes tissues, which were 1.23 ± 0.13 and 1.15 ± 0.09, respectively. The benign ovarian tissues were the second, (1.09 ± 0.11), while it was the lowest in paracancer tissues and normal ovarian tissues (0.32 ± 0.07,0.21 ± 0.04, P <0.05). Conclusion The expression of OPN is related to the malignant degree of ovarian cancer and may be related to the occurrence of ovarian cancer. The detection of OPN expression may be helpful to guide the clinical treatment and evaluate the prognosis of patients.
