目的:研究西红花苷对人胚肾细胞(human embryonic kidney 293cell,HEK293)上表达的延迟整流钾电流的作用。方法:运用脂质体转染法,将人心肌细胞上的延迟整流钾电流通道蛋白基因转染到HEK293细胞上,采用穿孔膜片钳法记录西红花苷对快激活延迟整流钾电流(The rapidly activating delayed rectifier potassium current,IKr)和慢激活延迟整流钾电流(The slowly activating delayed rectifier potassium current,IKs)的影响。结果:西红花苷对表达在HEK293上的IKr和IKs均无明显作用。结论:西红花苷不影响延迟整流钾电流,临床使用不易诱发QT间期延长和心律失常。
Objective: To study the effect of crocin on delayed rectifier potassium current expressed on human embryonic kidney 293cell (HEK293). Methods: The gene of delayed rectifier potassium channel in human cardiomyocytes was transfected into HEK293 cells by lipofection. The effects of crocin on delayed activation of delayed rectifier potassium current (IKr) and the slow activating delayed rectifier potassium current (IKs). Results: Crocin had no effect on IKr and IKs expressed on HEK293. Conclusion: Crocetin does not affect the delayed rectifier potassium current, clinical use is not easy to induce QT prolongation and arrhythmia.
