目的观察雷公藤多苷(TWP)对糖尿病肾病(DN)大鼠肾脏结缔组织生长因子(CTGF)表达和排泄的影响。方法用腹腔注射链脲佐菌素的方法建立糖尿病大鼠模型。动物随机分为正常组、模型组、3个剂量(45,90,180mg·kg-1)TWP组;用药8周末,检测大鼠血糖(BG)、体重(BW)、肾重(KW)、肾重/体重(KW/BW)B、24 h尿微量蛋白清除率(UAER)、血尿素氮(BUN)和肌酐清除率(Ccr);HE染色观察大鼠肾组织病理学变化;免疫组化检测肾皮质CTGF蛋白表达。结果与模型组相比,TWP能明显改善DN大鼠一般情况,高剂量TWP组空腹BG明显降低(P<0.01),中、高剂量TWP组能使BW明显增加(P<0.01),同时使KW、KW/BW、24hUAER、BUN及Ccr也明显降低(P<0.01),使肾组织着色程度明显减轻,使CTGF表达显著下降(P<0.01),并使肾小球肥大、系膜细胞增生及肾小球变形病变均明显减轻。结论 TWP能明显改善肾脏功能,且与剂量相关,其机制可能与抑制肾脏CTGF表达有关。
Objective To observe the effect of Tripterygium glycosides (TWP) on the expression and excretion of renal connective tissue growth factor (CTGF) in diabetic nephropathy rats. Methods The model of diabetic rats was established by intraperitoneal injection of streptozotocin. The animals were randomly divided into normal group, model group and three doses (45, 90 and 180 mg · kg -1) of TWP group. After 8 weeks of treatment, the blood glucose (BG), body weight (BW), kidney weight (W / BW) B, urine UAER, BUN and Ccr 24 h; pathological changes of renal tissue were observed by HE staining; immunohistochemistry Renal cortical CTGF protein expression. Results Compared with the model group, TWP significantly improved the general condition of DN rats. Conclusion TWP can significantly improve renal function, and dose-related, and its mechanism may be related to the inhibition of renal CTGF expression.
