目的探讨脱氧胆酰酪氨酸热敏凝胶剂(deoxycholyltyosine thermoreversible gel,DCT gel)对小型猪阴道黏膜的炎性细胞数量及活化状态的影响。方法将9只雌性小型猪随机分为基质对照组、壬苯醇醚胶冻剂(Nonxgnol-9gel,N-9 jelly)组和DCT gel组,肌注苯甲酸雌二醇12 d后开始阴道给药,每天1次连续给药7 d,在末次给药后24与72 h,收集阴道灌流(Cervicovaginal lavage,CVL)液,流式细胞仪计数炎性细胞和中性粒细胞数目,并对CD45+细胞进行表型分析,比较不同给药组之间的差别。结果 DCT gel组CVL液中炎性细胞、中性粒细胞数目以及CD45+细胞数目明显低于N-9 jelly组(P<0.001),与基质对照组比较差异无统计学意义(P>0.05)。结论脱氧胆酰酪氨酸热敏凝胶剂(DCT gel)不会引起小型猪阴道黏膜的明显刺激,未诱导局部炎性细胞的活化,作为外用杀精剂具有一定的临床应用前景。
Objective To investigate the effect of deoxycholyltyosine thermoreversible gel (DCT gel) on the number of inflammatory cells and the activation status of vaginal mucosa in miniature pigs. Methods Nine female miniature pigs were randomly divided into control group, nonxgnol-9 gel (N-9 jelly) group and DCT gel group. Intravenous estradiol benzoate was administered intraperitoneally The drug was administered once a day for 7 consecutive days. Cervicovaginal lavage (CVL) fluid was collected 24 and 72 h after the last administration. The number of inflammatory cells and neutrophils was counted by flow cytometry. Cells were phenotypically analyzed to compare the differences between the different dosing groups. Results The number of inflammatory cells, neutrophils and CD45 + cells in CVL fluid of DCT gel group was significantly lower than that in N-9 jelly group (P <0.001). There was no significant difference between CVT group and control group (P> 0.05). Conclusion DCT gel does not cause significant stimulation of vaginal mucosa in minipig and does not induce the activation of local inflammatory cells. It has some clinical application prospects as topical spermicide.
