目的:进行芹菜根降血脂作用有效部位的筛选,阐明其作用机制。方法:将芹菜根提取物及分离单体加入含有AH109菌株(携带大鼠apo-AⅠ与SR-BⅠ)的培养基中,通过报告基因ɑ,β-半乳糖苷酶活力的测定来定量检测apo-AⅠ与SR-BⅠ之间互相作用力的强弱,从而反应被测药物的降脂活力大小。结果:正丁醇组分中分离出的单体能够增强两种蛋白的相互作用。结论:芹菜根中正丁醇组分中分离出的这一单体是降血脂的目标化合物。
OBJECTIVE: To screen the effective parts of celery root hypolipidemic effect and clarify its mechanism. Methods: The celery root extract and the isolated monomer were added into the medium containing AH109 strain (carrying rat apo-AⅠ and SR-BⅠ), and the apo (apo) and β-galactosidase -A Ⅰ and SR-B Ⅰ interaction between the strength of the reaction of the measured drug lipid-lowering activity. Results: The monomer isolated from the n-butanol fraction enhances the interaction between the two proteins. CONCLUSION: This monomer isolated from the n-butanol fraction of celery root is the target compound for lipid lowering.
