目的:建立RP-HPLC法同时测定四季三黄软胶囊中黄芩苷和盐酸小檗碱的含量。方法:采用色谱柱为hypersil BDS C_(18)(250mm×4.6 mm,5μm),以乙睛-0.5%三乙胺水溶液(磷酸调pH至3.1)(25:75)为流动相,检测波长265 nm,流速1.0 ml·min~(-1)。结果:黄芩苷在1.6～206μg·ml~(-1)范围内,线性关系良好(r=0.999 9),平均回收率为100.3%。RSD=1.7%(n= 6);盐酸小檗碱在1.6～208μg·ml~(-1)范围内,线性关系良好(r=0.9999),平均回收率为100.8%,RSD=1.6%(n=6)。结论:该方法结果准确可靠,重现性好,可用于四季三黄软胶囊的质量控制。
Objective: To establish a RP-HPLC method for simultaneous determination of baicalin and berberine hydrochloride in Sijisanhuang soft capsules. Method: The chromatographic column was hypersil BDS C 18 (250 mm × 4.6 mm, 5 μm) with acetonitrile-0.5% aqueous solution of triethylamine (pH adjusted to 3.1 with phosphoric acid) (25:75) as the mobile phase. The detection wavelength was 265. Nm, flow rate 1.0 ml·min~(-1). RESULTS: The linear relationship between baicalin in the range of 1.6-206 μg·ml -1 was good (r=0.999 9) and the average recovery was 100.3%. RSD was 1.7% (n=6); berberine hydrochloride had a good linear relationship (r=0.9999) in the range of 1.6-208 μg·ml-1, with an average recovery of 100.8% and RSD=1.6% (n =6). Conclusion: The method is accurate, reliable and reproducible. It can be used for the quality control of Siji Sanhuang soft capsules.
