目的探讨在全反式维甲酸(ATRA)、1,25二羟维生素D3[1,25-(OH)2D3]的作用下,白血病细胞株SHI-1中多肽N-乙酰氨基半乳糖转移酶(ppGalNAcTs)和某些N-乙酰氨基葡萄糖转移酶(GnTs)表达的变化。方法应用RT-PCR法研究在不同浓度ATRA、1,25-(OH)2D3作用下,白血病细胞株SHI-1中ppGalNAcTs、GnTs表达的变化。结果在SHI-1细胞株中,随着1,25-(OH)2D3浓度(10-8~10-6mol/L)的增加,ppGalNAcT1和T2表达增加,T3表达没有变化,T4表达减少;加入ATRA(10-8~10-6mol/L)后,细胞株中ppGalNAcT2、β6GnTV、O-GnT表达量升高。结论白血病细胞株SHI-1ppGalNAcTs各型表达水平不同;在不同浓度ATRA、1,25-(OH)2D3的作用下,白血病细胞株SHI-1中各型ppGalNAcTs表达变化也不同。在SHI-1细胞株加入ATRA后细胞中β2GnTI、β6GnTV发生显著变化。ppGal-NAcT1、ppGalNAcT2、GnTs与白血病细胞株的分化可能有一定关系。
Objective To investigate the effect of N-acetylgalactosaminyltransferase (N-acetylgalactosamine transferase) on the expression of SHI-1 in leukemia cell line ATRA and 1,25-dihydroxyvitamin D3 [1,25- (OH) 2D3] ppGalNAcTs) and some N-acetylglucosaminyltransferase (GnTs) expression changes. Methods The expression of ppGalNAcTs and GnTs in leukemia cell line SHI-1 was studied by RT-PCR with different concentrations of ATRA, 1,25- (OH) 2D3. Results In SHI-1 cell line, the expression of ppGalNAcT1 and T2 increased with the increase of concentration of 1,25- (OH) 2D3 (10-8 ~ 10-6 mol / L), the expression of T3 did not change and the expression of T4 decreased. After ATRA (10-8 ~ 10-6mol / L), the expression of ppGalNAcT2, β6GnTV and O-GnT in cell lines increased. Conclusions The expression levels of various types of SHI-1ppGalNAcTs in leukemia cell line are different. The expression of various types of ppGalNAcTs in SHI-1 leukemia cell line is different under different concentrations of ATRA and 1,25- (OH) 2D3. After addition of ATRA in SHI-1 cell line, the changes of β2GnTI and β6GnTV in cells were significantly changed. ppGal-NAcT1, ppGalNAcT2, GnTs and leukemia cell lines may have a certain relationship.
