①目的探讨阿霉素诱导S-180瘤细胞凋亡过程中细胞核型的变化。②方法用低剂量(3.25μg)阿霉素处理经S-180瘤细胞腹腔接种的BALA/C鼠,0~72h分别取腹水进行细胞核型检测。③结果用阿霉素处理24h时,细胞变圆,胞浆浓缩;48h时,染色质密集度增高;72h时,核仁裂解,核膜内陷,形成内涵物不外溢的凋亡小体。④结论阿霉素是良好的细胞凋亡诱导剂,凋亡期间细胞外形及染色质凝聚逐渐改变,直至形成凋亡小体。
Objective To investigate the changes of karyotype in the process of apoptosis induced by doxorubicin in S-180 tumor cells. 2 Methods BALA/C mice inoculated intraperitoneally with S-180 tumor cells were treated with low dose (3.25μg) doxorubicin. Ascites were taken from 0 to 72h for karyotype detection. 3 Results When treated with doxorubicin for 24 hours, the cells became round and the cytoplasm was concentrated; at 48 hours, the chromatin density increased; at 72 hours, the nucleolus was lysed and the nuclear membrane retracted, forming apoptotic bodies with no inclusions. 4 Conclusions Doxorubicin is a good inducer of apoptosis, and cell morphology and chromatin condensation gradually change during apoptosis until the formation of apoptotic bodies.
