选取20份棉花品种,利用SSR分子标记法进行DNA指纹图谱的构建和遗传多样性分析。本研究从92对引物中筛选到15对核心引物,共检测到57种基因型,平均每对引物检测到的基因型数为3.8个,变幅为2~12;引物多态信息含量(PIC)值介于0.7308~0.9571,平均值为0.8782。结果表明,所选的15对引物中,TMB1152是10615-3、新陆早52、ND359-1、新陆早35、Y1169、新陆早36和新陆早47的特异性引物;BNL2646、CGR5565、NAU2238分别是ND359-1、ND359-2和08283、ND012以及10615-3、新陆早36的特异性引物;JESPR157、CIR332分别是新陆早48和Y1169的特异性引物,剩余品种利用引物组合法予以区分,成功建立了20份棉花品种的指纹图谱。
Twenty cotton cultivars were selected and the DNA fingerprinting (SSR) molecular markers were used to establish the genetic diversity analysis. In this study, 15 pairs of primers were screened from 92 pairs of primers. A total of 57 genotypes were detected, with an average of 3.8 genotypes per primer pair ranging from 2 to 12. The polymorphic information content (PIC ) Values ranged from 0.7308 to 0.9571 with an average of 0.8782. Among 15 selected primers, TMB1152 was the specific primer of 10615-3, Xinluzao 52, ND359-1, Xinluzao 35, Y1169, Xinluzao 36 and Xinluzao 47; BNL2646, CGR5565 , NAU2238 were ND359-1, ND359-2 and 08283, ND012 and 10615-3 respectively, and Xinluzao 36 specific primers. JESPR157 and CIR332 were specific primers of Xinluzao 48 and Y1169, respectively. France to be distinguished, the successful establishment of the fingerprint of 20 varieties of cotton.
