目的:研究甲型副伤寒沙门菌gyrA基因突变情况与喹诺酮药物敏感性降低的相关性。方法:通过微量稀释法测定甲型副伤寒沙门菌对萘啶酸和3种氟喹诺酮药物的敏感性,用PCR方法扩增其gyrA基因并进行序列分析,比较不同喹诺酮药物敏感性菌株的gyrA基因突变特点和差异。结果:gyrA基因的突变率和分布对不同喹诺酮药物敏感性的影响不同,特别是在gyrA基因的Ser83位点呈现明显差别。结论:甲型副伤寒沙门菌分离株gyrA基因突变与喹诺酮药物敏感性存在关联,可导致对喹诺酮耐药或敏感性下降。
Objective: To study the relationship between gyrA gene mutation of Salmonella paratyphi A and the decrease of quinolone sensitivity. Methods: The sensitivity of Salinomycin A to nalidixic acid and three fluoroquinolones drugs was determined by microdilution method. The gyrA gene was amplified by PCR and sequenced. The gyrA gene of different quinolone drug sensitive strains was compared Mutation characteristics and differences. Results: The mutation rate and distribution of gyrA gene had different effects on the sensitivity of different quinolones, especially the Ser83 of gyrA gene. Conclusion: The gyrA gene mutation of Salmonella paratyphi A is associated with quinolone sensitivity, which may result in the decrease of quinolone resistance or sensitivity.
