目的 比较SYBRGreenI染色及银染方法在定量聚合酶链反应 (Q PCR)诊断 2 1 三体综合征 (2 1 三体 )中的优劣。方法 PCR扩增 2 6例 2 1 三体及 2 0例正常人DNA ,分别用琼脂糖电泳 SYBRGreenI染色及聚丙烯酰胺凝胶 银染方法检测 ,进行光密度定量分析及比较。同时以细胞遗传学核型分析作对照。结果 两种方法检测DNA均于 2 4个PCR循环时显带清晰 ;定量分析结果同核型分析结果一致 ;SYBRGreenI染色历时30min,而银染则历时 4~ 5h。结论 在Q PCR中SYBRGreenI染色敏感度同银染 ,较银染更简单、省时、经济
Objective To compare the advantages and disadvantages of SYBRGreenI staining and silver staining in the diagnosis of trisomy 21 (2 1 trisomy) by quantitative polymerase chain reaction (Q PCR). Methods DNA of 26 cases with 21 trisomy and 20 normal individuals were amplified by PCR. The cells were stained with SYBR Green Ⅰ agarose gel electrophoresis and polyacrylamide gel silver staining respectively. The optical density was quantitatively analyzed and compared. At the same time cytogenetic karyotype analysis as a control. Results The DNA of the two methods were clearly banding in 24 PCR cycles. The results of quantitative analysis were consistent with the results of karyotype analysis. SYBRGreenI staining took 30min, while silver staining lasted 4 ~ 5h. Conclusion The sensitivity of SYBRGreenI staining in Q PCR is the same as silver staining, which is simpler than silver staining, saving time and economy
