目的:建立HPLC法同时测定五指毛桃中槲皮素、柚皮素、木犀草素、芹菜素、金合欢素、山柰酚6个成分的含量.方法:采用Syncronis C18色谱柱(250 mm ×4.6 mm,5μm);流动相为甲醇-0.2%磷酸水,梯度洗脱((0~10 min,55%B;10~15 min,55%B→100%B;15~20 min,100%B));检测波长320 nm;流速1.0mL·min-1;柱温30 ℃;进样量10μL.结果:将实验测得的图谱与标准物质对照,确定其6个成分,分别为槲皮素、柚皮素、木犀草素、芹菜素、金合欢素和山柰酚;6个成分分别在测定范围内(0.072~1.152μg,0.054~0.864 μg,0.062~0.992 μg,0.031~0.496 μg,0.068~1.088 μg,0.049~0.784 μg)线性关系良好;平均加样回收率为98.2%~100.2%,RSD均小于3.0%;10批样品中6个成分的含量分别为0.015%~0.028%、0.011%~0.020%、0.013%~0.031%、0.010%~0.022%、0.017%~0.059%和0.012%~0.039%.结论:该方法简便准确,重复性好,可用于五指毛桃药材6个成分的含量测定.
