目的探讨p33ING1对pcDNA3.1-p33ING1转染后的HepG2肝癌细胞p21WAF1/CIP1表达的影响。方法实验分为对照组和转染组;将pcDNA3.1-p33ING1及空载质粒pcDNA3.1转染到HepG2细胞中,最终建立稳定转染的细胞株,RT-PCR检测p33ING1表达;RT-PCR和免疫组化检测P21WAF1/CIP1mRNA和蛋白质表达。结果与对照组比较,转染组p33ING1mRNA表达明显增高(P<0.05),p21WAF1/CIP1mRNA及蛋白质表达亦明显高于对照组(P<0.05)。结论成功建立pcDNA3.1-p33ING1的HepG2细胞株,并证实p33ING1影响p21WAF1/CIP1mRNA和蛋白质的表达。
Objective To investigate the effect of p33ING1 on p21WAF1 / CIP1 expression in HepG2 hepatoma cells transfected with pcDNA3.1-p33ING1. Methods The experiment was divided into control group and transfection group. PcDNA3.1-p33ING1 and pcDNA3.1 were transfected into HepG2 cells, and stable transfected cell lines were finally established. The expression of p33ING1 was detected by RT-PCR. RT-PCR And immunohistochemical detection of P21WAF1 / CIP1 mRNA and protein expression. Results Compared with the control group, the expression of p33ING1 mRNA in the transfection group was significantly increased (P <0.05), and the expression of p21WAF1 / CIP1 mRNA and protein in the transfected group was significantly higher than that in the control group (P <0.05). Conclusion The pcDNA3.1-p33ING1 HepG2 cell line was successfully established and confirmed that p33ING1 affects the expression of p21WAF1 / CIP1 mRNA and protein.
