人源化CD52单克隆抗体诱导食蟹猴动物模型的建立

目的:建立人源化CD52单克隆抗体诱导食蟹猴的动物模型。方法:使用流式细胞仪筛选出红细胞表面不表达CD52抗原的食蟹猴,并观察给药后不同时间血中淋巴细胞数量变化情况。结果:食蟹猴血中淋巴细胞呈现出快速被清除及恢复的过程。CD20+B淋巴细胞、CD8+T淋巴细胞、CD4+T淋巴细胞分别在给药后213、5和56 d恢复至给药前水平。结论:食蟹猴外周血中淋巴细胞清除效果略低于临床病人,恢复速度较快。本实验模型可用于临床人源化CD52单克隆抗体器官移植前的实验研究。 Objective: To establish an animal model of cynomolgus monkeys induced by humanized CD52 monoclonal antibody. Methods: The cynomolgus monkeys that did not express CD52 antigen on the surface of erythrocytes were screened by flow cytometry. The changes of the number of lymphocytes in the blood were observed at different time points after administration. Results: The blood lymphocytes in cynomolgus monkey showed the process of rapid clearance and recovery. CD20 + B lymphocytes, CD8 + T lymphocytes and CD4 + T lymphocytes returned to pre-dose level at 213, 5 and 56 d after administration, respectively. Conclusion: Lymphocyte clearance in peripheral blood of cynomolgus monkeys is slightly lower than that of clinical patients, and the recovery rate is faster. The experimental model can be used for clinical study of humanized CD52 monoclonal antibody before organ transplantation.
