目的观察肝细胞肝癌(HCC)患者外周血单个核细胞(PBMC)中程序性死亡蛋白1(PD-1)和配体以及γ干扰素(IFN-γ)水平的变化。方法采集15例早期HCC患者、13例进展期HCC患者和12例健康人(对照组)的外周血并分离PBMC。流式细胞术检测PBMC中PD-1、PD-L1和PD-L2的表达,ELISA检测血清中IFN-γ的水平,利用Pearson相关系数法和One-way ANOVA分析PD-1与IFN-γ的相关性。结果进展期HCC患者外周血CD3+T细胞和CD19+B细胞中PD-1、PD-L1和PD-L2的表达水平和血清IFN-γ水平显著高于健康人群和早期HCC患者,HCC患者外周血CD3+T细胞和CD19+B细胞PD-1、PD-L1表达水平与IFN-γ呈正相关。结论肝癌患者PBMC中PD-1、PD-L1和PD-L2分子表达增加,且PD-1、PD-L1与IFN-γ水平有关联。
Objective To observe the changes of PD-1, ligand and IFN-γ in peripheral blood mononuclear cells (PBMCs) from patients with hepatocellular carcinoma (HCC). Methods Peripheral blood samples from 15 patients with early HCC, 13 patients with advanced HCC and 12 healthy controls (control group) were collected and PBMCs were isolated. The expression of PD-1, PD-L1 and PD-L2 in PBMCs were detected by flow cytometry. The level of IFN-γ in serum was detected by ELISA. The Pearson correlation coefficient and One-way ANOVA were used to analyze the expression of PD-1 and IFN- Correlation. Results The expression of PD-1, PD-L1 and PD-L2 and the level of serum IFN-γ in peripheral blood CD3 + T cells and CD19 + B cells in advanced HCC patients were significantly higher than those in healthy and early HCC patients The levels of PD-1 and PD-L1 in CD3 + T cells and CD19 + B cells were positively correlated with IFN-γ. Conclusion The expression of PD-1, PD-L1 and PD-L2 in PBMC of hepatocellular carcinoma patients is increased, and the levels of PD-1, PD-L1 and IFN-γ are correlated.
