目的:探讨骨碎补总黄酮含药血清对大鼠成骨细胞中雄激素受体(androgen receptor,AR)mRNA表达的影响。方法:取1日龄SD大鼠分离原代成骨细胞培养后传代,进行细胞鉴定。细胞分为3组,分别进行骨碎补总黄酮含药血清、丙酸睾酮、空白血清干预,RT-PCR方法测定成骨细胞雄激素受体mRNA表达的结果。结果:雄激素干预组及骨碎补干预组的AR mRNA表达率差异无统计学意义(P>0.05),表明骨碎补总黄酮的效应类似于丙酸睾酮。结论:骨碎补总黄酮可促进AR mRNA表达,可能与拟雄激素效应有关,从而可抑制骨吸收和促进骨生成,最终达到治疗雄激素水平下降所致骨质疏松症的目的。
Objective: To investigate the effects of total flavonoids of Rhizoma Drynariae on the mRNA expression of androgen receptor (AR) in rat osteoblasts. Methods: Primary osteoblasts isolated from 1-day-old SD rats were cultured and passaged for cell identification. The cells were divided into three groups. The serum containing total flavonoids of Osteopractic and Osteoporosis, testosterone propionate and blank serum were respectively used to detect the expression of androgen receptor mRNA in osteoblasts by RT-PCR. Results: There was no significant difference in AR mRNA expression between Andrographolide group and Rhizoma Drynariae group (P> 0.05), indicating that the effect of total flavonoids of Rhizoma Drynariae was similar to that of testosterone propionate. Conclusion: Total Flavonoids of Rhizoma Drynariae can promote the expression of AR mRNA, which may be related to androgen-induced effect, which can inhibit bone resorption and promote osteogenesis, and eventually achieve the purpose of treating osteoporosis caused by decreased androgen levels.
