目的探讨促凋亡基因PTEN Bi m蛋白在子宫内膜癌发生发展中的作用。方法应用免疫组化SP方法和Western blot方法检测17例正常增殖期子宫内膜、11例非典型增生子宫内膜及52例子宫内膜癌中PTEN、Bi m蛋白的表达。结果Bi m在正常子宫内膜中阳性表达率为100%,而在非典型增生内膜中阳性表达率开始下降(71.4%),在子宫内膜癌组织中明显下降,(P<0.05),而在子宫内膜癌各组织分级、临床分期及肌层浸润程度之间Bi m蛋白表达阳性率均无明显差异(P>0.05)。正常增殖期子宫内膜组织中PTEN表达量最多,随着内膜细胞的增殖程度的增加,PTEN表达量逐渐下降。结论PTEN Bi m蛋白表达改变与子宫内膜癌的发生密切相关。
Objective To investigate the role of pro-apoptotic gene PTEN Bi m in the development of endometrial carcinoma. Methods The expressions of PTEN and Bi m in 17 cases of normal proliferative endometrium, 11 cases of atypical hyperplasia and 52 cases of endometrial carcinoma were detected by immunohistochemical SP method and Western blot. Results The positive expression rate of Bi m in normal endometrium was 100%, while the positive expression rate in atypical hyperplasia endometrium began to decrease (71.4%), which was significantly decreased in endometrial carcinoma (P <0.05) However, there was no significant difference in the positive rate of Bi m protein between the histological grade, clinical stage and myometrial invasion in endometrial carcinoma (P> 0.05). The expression of PTEN in normal proliferative phase of endometrium was the highest, with the proliferation of endometrial cells increasing, the expression of PTEN gradually decreased. Conclusion The change of PTEN Bi m protein expression is closely related to the occurrence of endometrial carcinoma.
