用明胶和戊二醛将乳酸脱氨酶(LDH)固定于PVC颗粒表面后,装成酶柱,以流通池光纤荧光光度法连续测定乳酸,检测乳酸的线性范围为2.0×10-5~4.0×10-1mol·L-1。该方法简便、快速,重现性、稳定性均好,酶的使用寿命长,已用于酿牛奶和食用白醋中乳酸的测定。
Lactate deaminase (LDH) was immobilized on the surface of PVC particles with gelatin and glutaraldehyde, and then loaded into enzyme column. The concentration of lactic acid in the flow cell was determined by fluorescence spectrophotometry. The linear range was 2.0 × 10-5 ~ 4.0 × 10-1mol·L-1. The method is simple, rapid, reproducible, stable and good, long life of the enzyme, has been used for determination of lactic acid in milk and edible white vinegar.
