目的和方法 :给予健康雄性小鼠 (Balb/C)小柴胡汤 (TSS) ,糖皮质激素 (GC) ,应用受体放射配体结合分析法测定小鼠脾细胞糖皮质激素受体 (GR)位点数 ,同时用定量逆转录 多聚酶链反应 (RT PCR)法测定脾细胞内GRmRNA水平 ,以期从分子受体水平和基因水平探讨TSS对GR及GRmRNA的调节作用。结果和结论 :(1)应用TSS后GR位点数及GRmRNA水平明显少于正常对照组 ,说明TSS对GR的降调是通过调节GRmRNA水平实现的 ;(2 )TSS与GC合用时GR位点数及GRmRNA水平明显高于单独应用GC组 ,说明TSS可明显减弱GC对GR的降调作用。
Aims and methods: Healthy male mice (Balb/C) Xiao Chai Hu Tang (TSS) and glucocorticoid (GC) were used to determine splenocyte glucocorticoid receptor (GR) in mouse spleen using receptor ligand binding assay. At the same time, the level of GR mRNA in spleen cells was determined by quantitative reverse transcriptase polymerase chain reaction (RT PCR) method in order to investigate the regulatory effects of TSS on GR and GR mRNA levels from the molecular receptor level and gene level. RESULTS AND CONCLUSION: (1) The number of GR sites and GR mRNA levels after TSS was significantly lower than that of the normal control group, indicating that the down-regulation of GR by TSS was achieved by adjusting the GR mRNA levels; (2) the number of GR sites when TSS and GC were combined GR mRNA levels were significantly higher than those of GC alone, indicating that TSS significantly attenuated the down-regulation of GR by GC.
