目的寻找到适宜做影像学和血清学研究的动物家兔肝纤维化模型建立方法。方法分别用体积比为5%四氯化碳(CCl4)油溶液和纯CCl4,按0.1 ml/kg剂量每周1次腹腔注射,并辅以5%浓度的乙醇作为饮水,诱导家兔肝纤维化形成,并于注射后6、8、10、12周分组处死家兔取其肝脏进行病理学检查。结果家兔的死亡多发生在4周之内,6周之后趋于稳定。5%CCl4浓度组家兔死亡率为60%,纯CCl4组死亡率为25%。结论长期给予CCl4可导致家兔的肝纤维化形成,采用腹腔注射纯CCl,同时辅以5%浓度的乙醇作为饮水的复合法造模,家兔死亡率低,成模率高。
OBJECTIVE: To find a suitable animal model of rabbit hepatic fibrosis for imaging and serology. Methods The intraperitoneal injection of 5% Cetyl alcohol (CCl4) solution and pure CCl4 in the volume of 0.1 ml / kg and the ethanol in the concentration of 5% were used to induce the hepatic fibrosis in rabbits At 6, 8, 10, and 12 weeks after injection, rabbits were killed and their livers were taken for pathological examination. The results of the death of rabbits occurred in more than 4 weeks, after 6 weeks tends to be stable. The mortality rate of rabbits in 5% CCl4 group was 60% and that in pure CCl4 group was 25%. Conclusion Chronic administration of CCl4 can lead to the formation of hepatic fibrosis in rabbits. The intraperitoneal injection of pure CCl and supplemented with 5% ethanol as drinking water make the model of rabbits with low mortality rate and high rate of mold formation.
