目的了解中期因子 (Midkine,MK)转录物及其蛋白产物在肝细胞癌 (hepatocellularcarci noma ,HCC)中的定位情况与表达特点。方法应用原位杂交、免疫组织化学染色等方法对 33例人HCC组织、10例良性肝肿瘤组织及其配对瘤旁肝组织进行了MKmRNA及蛋白的定位与表达研究。结果免疫组织化学结果与原位杂交结果具有一致性 (χ2 =0 5 0 0 ,P >0 0 5 )。MK在HCC组织中呈高表达。MKmRNA及蛋白的阳性信号聚集于HCC细胞质。在HCC细胞外组织中亦有MK表达 ,尤以血管密集处明显。HCCMK表达率与肝癌组织学类型、分级等临床病理学特点无相关性。结论HCC在mRNA和蛋白水平上表达MK增加 ,并可能与促进HCC血管生成有关
Objective To investigate the localization and expression of midkine (MK) transcripts and their protein products in hepatocellular carcinoma (HCC). Methods The localization and expression of MK mRNA and protein in 33 cases of HCC, 10 cases of benign liver neoplasm and paired para-tumorous liver tissues were studied by in situ hybridization and immunohistochemical staining. Results The results of immunohistochemistry were consistent with those of in situ hybridization (χ2 = 0500, P> 0.05). MK is highly expressed in HCC tissues. Positive signals of MK mRNA and protein accumulate in the cytoplasm of HCC. In HCC extracellular tissue also has MK expression, especially in vascularized areas. HCCMK expression rate and histological type of liver cancer, grade and other clinical pathological features without correlation. Conclusion The expression of HCC at mRNA and protein level increased, which may be related to the promotion of HCC angiogenesis
