目的探讨心肌梗死患者microRNA-21(miR-21)和转化生长因子(TGF)-β1的表达水平,以及调控心肌梗死后心室重构发生的作用机制。方法选取200例临床和心电图确诊为心肌梗死患者血浆和100例健康体检人群血浆,分别采用荧光定量PCR检测各组血浆miR-21和TGF-β1的表达水平。结果与健康对照组比较,心肌梗死患者血浆miR-21的表达在心肌梗死后第3天、第7天、第14天,分别为0.74±0.21 vs.2.62±0.23,vs.3.67±0.25,vs.4.13±0.27,P<0.05;心肌梗死患者血浆TGF-β1的表达在心肌梗死后第3天、第7天、第14天,分别为0.98±0.18 vs.2.35±0.24,vs.3.67±0.25,vs.4.13±0.27,P<0.05。心肌梗死患者血浆miR-21和TGF-β1随着心肌梗死后心功能变化表达上调。血浆miR-21和TGF-β1mRNA与左心室舒张末内径呈正相关(r=0.757、0.701,P<0.05)。结论心肌梗死患者血浆miR-21和TGF-β1表达上调,miR-21和TGF-β参与调控心肌梗死后的心室重构。
Objective To investigate the expression of microRNA-21 (miR-21) and transforming growth factor-β1 (TGF-β1) in myocardial infarction patients and to study the mechanism of ventricular remodeling after myocardial infarction. Methods Plasma samples of 200 patients with myocardial infarction and 100 healthy people diagnosed by clinical and electrocardiogram were selected and their plasma levels of miR-21 and TGF-β1 were detected by real-time PCR. Results Compared with healthy control group, the expression of plasma miR-21 in myocardial infarction patients were 0.74 ± 0.21 vs.2.62 ± 0.23 vs.3.67 ± 0.25 on the 3rd, 7th and 14th day after myocardial infarction, vs .4.13 ± 0.27, P <0.05; The expression of TGF-β1 in myocardial infarction patients was 0.98 ± 0.18 vs.2.35 ± 0.24, vs.3.67 ± 0.25 on the 3rd, 7th and 14th day after myocardial infarction , vs.4.13 ± 0.27, P <0.05. The levels of plasma miR-21 and TGF-β1 in patients with myocardial infarction were up-regulated after cardiac infarction. Plasma miR-21 and TGF-β1 mRNA were positively correlated with left ventricular end-diastolic diameter (r = 0.757, 0.701, P <0.05). Conclusions The expression of miR-21 and TGF-β1 in plasma is increased in patients with myocardial infarction. MiR-21 and TGF-β are involved in the regulation of ventricular remodeling after myocardial infarction.
