目的采用大孔吸附树脂法,优化芪天滴丸中赤芍、红花及红景天活性成分的纯化工艺条件。方法以芍药苷、羟基红花黄色素A(HSYA)及红景天苷为指标,对4种树脂进行了筛选;以芍药苷和HSYA为指标,优化HPD100型大孔树脂富集纯化的工艺条件。结果HPD100型对芪天滴丸中芍药苷、HSYA及红景天苷具有很好的吸附特性,优化HPD100型大孔树脂富集纯化的工艺条件为:样品溶液含芍药苷2.98 g.L-1,含HSYA 1.44 g.L-1,含红景天苷0.81 g.L-1,吸附时间不低于12 h,吸附速度60 mL.h-1,洗脱速度60 mL.h-1,20%乙醇洗脱200 mL。大孔吸附树脂反复使用7次。结论HPD100大孔吸附树脂能有效富集纯化赤芍、红花及红景天中的活性成分。
Objective To purify the active components of Radix Paeoniae Rubra, Rhodiola and Rhodiola by using macroporous adsorption resin method. Methods Paeoniflorin, hydroxysafflor yellow A (HSYA) and salidroside were used as indexes to screen four kinds of resins. Paeoniflorin and HSYA were used as indexes to optimize the process conditions of HPD100 macroporous resin enrichment and purification . Results HPD100 had good adsorption properties for paeoniflorin, HSYA and salidroside in Qitian dropping pills. The optimal conditions for enrichment and purification of HPD100 macroporous resin were as follows: sample solution containing paeoniflorin 2.98 g L-1, HSYA 1.44 gL-1, containing salidroside 0.81 gL-1, the adsorption time of not less than 12 h, the adsorption rate of 60 mL.h-1, the elution rate of 60 mL.h-1, 20% ethanol 200 mL . Macroporous resin used repeatedly 7 times. Conclusion HPD100 macroporous adsorption resin can effectively enrich the purified active ingredients of Radix Paeoniae, Rhodiola and Rhodiola.
