目的探讨MDM2蛋白、p53在肺腺癌中的表达及其在肺腺癌发生发展中的作用。方法应用免疫组化技术检测41例肺腺癌组织中MDM2、P53的表达。结果MDM2、p53在肺腺癌中的表达率分别为31.71%(13/41),53.66%(22/41);原发灶≥3.0cm者,p53表达率为33.43%±7.28%,高于<3.0cm者6.61%±1.34%,(P<0.01);原发灶≥3.0cm者,MDM2表达率为3.77%±1.13%,低于<3.0cm者24.53%±4.13%(P<0.05)。MDM2表达与突变型P53表达呈负相关性,Sperman等级相关r=-0.54,P<0.05。两者均与肺腺癌病理分型相关,标准回归系数(Beta)值分别为:MDM20.443,p530.74。结论MDM2的表达与P53的表达存在负相关。p53/MDM2存在负反馈机制,是肺腺癌发生发展的分子基础,此外,p53MDM2各自还存在独立的作用机制。
Objective To investigate the expression of MDM2 protein and p53 in lung adenocarcinoma and its role in the development of lung adenocarcinoma. Methods Immunohistochemistry was used to detect the expression of MDM2 and P53 in 41 cases of lung adenocarcinoma. Results The positive rates of MDM2 and p53 in lung adenocarcinoma were 31.71% (13/41) and 53.66% (22/41), respectively. The positive rate of p53 was 33.43% ± 7.28% (P <0.01). The positive rate of MDM2 was 3.77% ± 1.13% in primary tumors ≥3.0cm, and 24.53% ± 4.13% (P <0.05) in those less than 3.0cm, . MDM2 expression was negatively correlated with mutant P53 expression, Sperman rank correlation was r = -0.54, P <0.05. Both were related to the pathological type of lung adenocarcinoma, and the standard values of Beta were MDM20.443 and p530.74, respectively. Conclusion The expression of MDM2 is negatively correlated with the expression of P53. The negative feedback mechanism of p53 / MDM2 is the molecular basis of the development of lung adenocarcinoma. In addition, there exists an independent mechanism of action of p53 / MDM2.
