用酶消化法获得大鼠皮肤肥大细胞,培养于含10%胎牛血清的DMEM培养液。培养1d后,在培养液中加入不同浓度的SP、SP+Ca2+或ConA,继续培养不同时间。用微孔滤膜迅速分离细胞和培养液,荧光法测定培养液中的组胺含量,计算组胺释放率。结果显示,SP和ConA均能以时间和剂量依赖性方式刺激体外培养的大鼠皮肤肥大细胞释放组胺,并且SP的这种作用明显地受Ca2+浓度的影响。
Rat skin mast cells were obtained by enzymatic digestion and cultured in DMEM medium containing 10% fetal bovine serum. After culturing for 1 day, different concentrations of SP, SP + Ca2 + or ConA were added into the culture medium and cultured for different time. The cells and culture fluid were rapidly separated using a microporous membrane filter, and the histamine content in the culture fluid was measured by fluorescence method to calculate the histamine release rate. The results showed that both SP and ConA stimulate histamine release from rat skin mast cells cultured in vitro in a time-and dose-dependent manner, and this effect of SP is significantly affected by Ca2 + concentration.
