目的研究+Gz暴露对冠脉不同程度狭窄模型猪血浆及血管巨噬细胞移动抑制因子(MIF)表达的影响。方法 25只巴马小型猪,在胸腔镜直视下手术丝线永久性结扎左前降支近端建立不同冠脉狭窄模型。各组模型猪进行+Gz暴露,并观察其最大+Gz耐受值。在最大+Gz暴露处理前后1 min留取静脉血并分离血浆,应用ELISA法检测MIF的含量。采用RT-PCR和免疫组化检测+Gz暴露后各组小型猪升主动脉MIF的表达情况。结果最大可耐受+Gz暴露后,各组模型猪MIF表达水平均较暴露前明显升高(P<0.05),且中、重度狭窄组模型猪血浆MIF浓度及升主动脉MIF表达水平均较假手术组明显升高(P<0.05),而轻度狭窄组则升高不明显(P>0.05)。结论 +Gz暴露对冠脉不同程度狭窄模型猪MIF的表达存在影响,且随狭窄程度加重,MIF表达增多。
Objective To investigate the effects of + Gz exposure on the expression of MIF in plasma and vascular of pigs with different degree of coronary artery stenosis. Methods Twenty-five Bama miniature pigs underwent thoracoscopic surgery to permanently ligate the proximal left anterior descending artery to establish different coronary artery stenosis models. The pigs in each group were exposed to + Gz and the maximum + Gz tolerance was observed. Venous blood was collected 1 min before and after the maximum + Gz exposure and plasma was separated. The content of MIF was measured by ELISA. The expression of MIF in ascending aorta of mini-pigs was detected by RT-PCR and immunohistochemistry after + Gz exposure. Results After M + Gz exposure, the expression of MIF in each model group was significantly higher than that before exposure (P <0.05), and the levels of MIF and the level of MIF in ascending aorta in moderate and severe stenosis group The sham operation group was significantly increased (P <0.05), while the mild stenosis group was not significantly increased (P> 0.05). Conclusions + Gz exposure affects the expression of MIF in pigs with varying degrees of coronary stenosis. MIF expression increases with the severity of stenosis.
