目的：探讨α－干扰素（ＩＦＮ－α）对人肺腺癌细胞株Ａ５４９细胞周期和Ｐ５３蛋白表达的影响。方法：用２种不 同质量浓度ＩＦＮ－α（２５μｇ／Ｌ，１００ μｇ／Ｌ）处理人肺腺癌Ａ５４９细胞，并设空白为对照。用流式细胞仪测定ＩＦＮ－α对Ａ５４９细 胞细胞周期的影响，用免疫荧光标记法测定Ｐ５３蛋白的表达。结果：ＩＦＮ－α作用后的Ａ５４９细胞，细胞周期明显改变，Ｓ期细 胞数显著降低，且Ｐ５３蛋白表达明显减少。结论： ＩＦＮ－α能阻止Ａ５４９细胞细胞周期的进程，抑制Ｐ５３蛋白的表达。
Objective: To investigate the effects of IFN-α on cell cycle and P53 protein expression in human lung adenocarcinoma cell line A549. Methods: Human lung adenocarcinoma A549 cells were treated with 2 different concentrations of IFN-α (25 μg / L, 100 μg / L) and blank was set as the control. The effect of IFN-α on the cell cycle of A549 cells was determined by flow cytometry. The expression of P53 protein was detected by immunofluorescence staining. Results: After A549 cells were treated with IFN-α, the cell cycle was significantly changed, the number of S phase cells was significantly decreased, and the expression of P53 protein was significantly decreased. Conclusion: IFN-α can inhibit the cell cycle progression of A549 cells and inhibit the expression of P53 protein.
