目的:探讨宫颈病变组织中核因子NF-κBp65、磷酸化型NF-κBp65(p-NF-κBp65)的表达及临床意义,以及高危型人乳头状瘤病毒(hsHPV)对其表达的影响。方法:采用免疫组化SP法检测NF-κBp65、p-NF-κBp65在67例宫颈癌、149例宫颈上皮内瘤样病变(CINⅠ-Ⅲ)、15例正常宫颈组织中的表达。结果:NF-κBp65、p-NF-κBp65的表达随宫颈病变级别升高而升高,并与宫颈病变级别呈正相关(P<0.05);NF-κBp65、p-NF-κBp65在hs-HPV阳性组的表达高于hs-HPV阴性组(P<0.05);NF-κBp65、p-NF-κBp65在癌细胞高分化、肿瘤直径≤4 cm、临床Ⅰ-Ⅱ期组的表达低于癌细胞中低分化、肿瘤直径>4 cm、临床Ⅲ-Ⅳ期组(P<0.05)。结论:hs-HPV能促进NF-κBp65的表达和激活,NF-κBp65的表达和激活可能与宫颈癌的发生、发展密切相关。
Objective: To investigate the expression and clinical significance of nuclear factor NF-κBp65 and phosphorylated NF-κBp65 in cervical lesions and the effect of high-risk human papillomavirus (hsHPV) on its expression. Methods: The expressions of NF-κBp65 and p-NF-κBp65 in 67 cases of cervical cancer, 149 cases of cervical intraepithelial neoplasia (CINⅠ-Ⅲ) and 15 cases of normal cervical tissue were detected by immunohistochemical SP method. Results: The expression of NF-κBp65 and p-NF-κBp65 increased with the increase of cervical lesions and positively correlated with the grade of cervical lesions (P <0.05). The positive expression of NF-κBp65 and p-NF- (P <0.05). NF-κBp65 and p-NF-κBp65 were highly differentiated in cancer cells with a diameter of less than 4 cm and the expression in clinical stage Ⅰ-Ⅱ was lower than that in hs-HPV negative group Poorly differentiated, tumor diameter> 4 cm, clinical stage Ⅲ-Ⅳ group (P <0.05). Conclusion: hs-HPV can promote the expression and activation of NF-κBp65. The expression and activation of NF-κBp65 may be closely related to the occurrence and development of cervical cancer.
