银杏平颤方对帕金森病鼠多巴胺神经元丢失和细胞凋亡的影响

目的探讨银杏平颤方防治帕金森病的机制。方法采用腹腔注射MPTP建立小鼠帕金森病模型。分别应用免疫组化方法和末端脱氧核苷酸转移酶介导的dUTP缺口末端标记法观察中药对中脑黑质多巴胺神经元丢失和细胞凋亡的影响。结果模型组小鼠中脑黑质致密部酪氨酸羟化酶阳性神经元的数目比正常组明显减少,28天比14天减少更明显;银杏平颤方及其拆方各中药治疗组可部分减少黑质致密部TH阳性神经元的丢失;模型组小鼠中脑黑质致密部凋亡细胞的数量明显增多,14天比28天增加更明显,中药各治疗组可不同程度地抑制该部位细胞的凋亡。结论细胞凋亡可能是帕金森病小鼠中脑多巴胺神经元丢失的主要方式之一,银杏平颤方可通过抑制细胞凋亡减少多巴胺神经元的丢失,达到防治帕金森病的作用。 Objective To explore the mechanism of prevention and treatment of Parkinson’s disease by ginkgo Pingqiao. Methods The mouse model of Parkinson’s disease was established by intraperitoneal injection of MPTP. Immunohistochemistry and terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling were used to observe the effects of Chinese herbs on dopamine neuron loss and apoptosis in the substantia nigra. Results The number of tyrosine hydroxylase-positive neurons in the substantia nigra pars compacta in the model group was significantly lower than that in the normal group, and it was more obvious on the 28th day than on the 14th day. The ginkgo Pingqiao Decoction and its deconstructed Chinese medicine treatment groups were available. The loss of TH-positive neurons in the substantia nigra pars compacta was partially reduced; the number of apoptotic cells in the substantia nigra pars compacta increased significantly in the model group, increasing more markedly in the 14th day than in the 28th day, and the treatment groups in the Chinese medicine could inhibit this to varying degrees. Apoptosis of the site cells. Conclusion Apoptosis may be one of the major ways of neuronal loss of midbrain dopaminergic neurons in mice with Parkinson’s disease. Ginkgo biloba flat fibroids can reduce the loss of dopaminergic neurons by inhibiting apoptosis and achieve the effect of preventing Parkinson’s disease.
