SRAP是一种可检测基因可译框区域多态性的分子标记系统,具有简便、稳定、易测序等优点。本研究利用SRAP标记对16份浙江柚类地方资源及2份柚与其他柑橘的杂交后代进行了遗传多样性研究。结果显示,49对引物中筛选出13对多态性引物,引物组合扩增条带数在2~14条,平均为9条带,多态率为0~100%,平均为73.5%。种质间的相似系数从0.513到0.932,其中四季柚和红心四季柚的相似系数最高,而管南土柚和玉环柚为最低。聚类分析结果显示,样品在遗传相似系数0.738处分为四类,第一类包括13份样品,第二类包括管南红肉抛和管南土柚,第三类为皋泄香柚,第四类包括常山胡柚和槾文柑。研究结果表明,浙江柚类资源遗传背景复杂,大多数种质由外地引入并驯化而成。
SRAP is a molecular marker system that can detect the gene polymorphism of the translated region of the gene, which has the advantages of simplicity, stability and easy sequencing. In this study, SRAP markers were used to study the genetic diversity of 16 local pomelo resources and 2 hybrids of pomelo and other citrus cultivars. The results showed that 13 pairs of polymorphic primers were screened out from 49 pairs of primers. The number of amplified bands was 2-14, with an average of 9 bands. The polymorphism rate was 0-100% with an average of 73.5%. The similarity coefficient of germplasm ranged from 0.513 to 0.932, of which the similarity coefficient between Siji grapefruit and Hongxin Siji grapefruit was the highest, while that of Guannan Pomelo and Yuhuan pomelo was the lowest. The results of cluster analysis showed that the samples were divided into four categories at the genetic similarity coefficient of 0.738, the first category consisted of 13 samples, the second category consisted of Guannan red meat dumpling and Guannan soil pomelo, the third category was Gao lei pomelo, Four categories include Changshan Huyou and Mandarin orange. The results showed that the pomelo resources in Zhejiang have a complex genetic background. Most of the germplasm was introduced and domesticated from the outside.
