目的:研究JUN基因在胎儿骨髓间质干细胞(FMSCs)、成人骨髓间质干细胞(AMSCs)、F6s、SGCs及白血病细胞中的差异表达,寻找肿瘤和白血病的分子生物学特征。方法:分别提取FMSCs、AMSCs、F6s、SGCs及白血病单个核细胞的总RNA,逆转录cDNA,以cDNA为模板用PCR和Real-time PCR技术测定JUN基因表达水平。结果:AML、ALL组JUN表达的阳性率高于CML,但尚无统计学意义,且JUN基因在AMSCs、FMSCs、F6s、SGCs组的表达差异无显著性(P>0.05)。结论:JUN基因在MSCs转化肿瘤细胞过程中其表达水平的变化呈异质性,推测与瘤变所处的环境有关;JUN基因与白血病类型的关系尚需积累病例进一步研究。
OBJECTIVE: To study the differential expression of JUN gene in fetal bone marrow mesenchymal stem cells (FMSCs), adult bone marrow mesenchymal stem cells (AMSCs), F6s, SGCs and leukemia cells in order to find the molecular biological characteristics of tumor and leukemia. Methods: Total RNA was extracted from FMSCs, AMSCs, F6s, SGCs and leukemic mononuclear cells respectively, cDNA was reverse transcribed, and JUN gene expression levels were determined by PCR and Real-time PCR using cDNA as a template. Results: The positive rate of JUN expression in AML and ALL patients was higher than that in CML patients, but there was no significant difference in JUN expression between AMSCs, FMSCs, F6s and SGCs (P> 0.05). CONCLUSION: JUN gene is heterogeneous in the expression of MSCs in tumor cells. It is presumed that the JUN gene is related to the environment in which tumor is located. The relationship between JUN gene and leukemia type needs to be further studied.
