目的 研究凋亡调控基因 bcl- 2和 bax在 AL中的表达、相互关系和临床意义。方法 应用半定量逆转录 -聚合酶链反应 ( RT- PCR)和 Sorthernbolt技术检测 32例 AL患儿 bcl- 2和 bax m RNA水平及其比值。结果 AL 患儿骨髓细胞中 bcl- 2 m RNA的表达明显高于对照组 ( P<0 .0 1) ,bax的表达明显降低 ( P<0 .0 1)。分析 bcl- 2 / bax m RNA表达比值显示 ,高比值组 NR率显著高于低比值组 ( P<0 .0 1)。结论 AL的发病与凋亡调控基因 bcl- 2和 bax的异常表达有关。bcl-2和 bax m RNA及其比值可作为判断 AL预后及指导治疗的主要监测指标
Objective To study the expression, relationship and clinical significance of apoptosis-regulating genes bcl-2 and bax in AL. Methods The levels of bcl-2 and bax m RNA in 32 children with AL were detected by semiquantitative RT-PCR and Sorthernbolt. Results The expression of bcl-2 mRNA in bone marrow cells of AL patients was significantly higher than that in control group (P<0.01), and the expression of bax was significantly decreased (P<0.01). Analysis of bcl-2 / bax m RNA expression ratio showed that the ratio of NR in the high ratio group was significantly higher than that in the low ratio group (P < 0.01). Conclusion The onset of AL is related to the abnormal expression of apoptosis-regulating genes bcl-2 and bax. The bcl-2 and bax m RNA and their ratios can be used as the main monitoring indicators to judge the prognosis and treatment of AL.
