目的:研究齿叶白鹃梅总黄酮提取物在大鼠体内的药物代谢动力学特点。方法:建立大鼠血浆中黄酮类化合物在大鼠体内的代谢产物对羟基桂皮酸HPLC测定方法,考察大鼠口服齿叶白鹃梅总黄酮提取物0.083、0.167、0.333、0.500、0.667、1.0、2.0、4.0、6.0、8.0、12.0、24.0h后血药浓度变化,采用DAS 2.0数据处理软件计算药代动力学参数。结果:大鼠口服齿叶白鹃梅总黄酮提取物后,对羟基桂皮酸在大鼠体内的达峰时间T_(max)为0.618h,最大浓度C_(max)为27.881mg·L~(-1),吸收半衰期t_(1/2Ka)为0.189 h,分布半衰期t_(1/2α)为4.367h,消除半衰期t_(1/2β)为12.232h,且在4h左右时第二次达峰,出现双峰现象。结论:建立了对羟基桂皮酸血药浓度的测定方法,该方法专属性强,灵敏度高,可用于该药的体内定量分析。
Objective: To study the pharmacokinetics of total flavonoids extract from Rhizoma Imperatae in rats. Methods: HPLC method was established for the determination of hydroxycinnamic acid in rat plasma by HPLC. The total flavonoids extract of Rhododendron syriacus Oralis was 0.083,0.167,0.333,0.500,0.667,1.0,2.0, 4.0, 6.0, 8.0, 12.0, 24.0h plasma concentration changes, the use of DAS 2.0 data processing software to calculate pharmacokinetic parameters. Results: After oral administration of total flavonoids from Rhododendron officinale, the peak time of p-hydroxycinnamic acid in rats was 0.618h and the maximum concentration was 27.881mg · L -1. , The half-life of half-life t_ (1/2 Ka) was 0.189 h, the half-life of distribution t_ (1 / 2α) was 4.367h, the elimination half-life was 12.232h, and reached the second peak at about 4h. Peak phenomenon. Conclusion: A method for the determination of plasma concentration of hydroxycinnamic acid was established. The method is highly specific and sensitive and can be used for in vivo quantitative analysis of the drug.
