目的 通过研究正常子宫内膜和子宫内膜癌组织基因表达的变化 ,来探讨子宫内膜癌的发生机制。方法 应用cDNA微阵列技术 (含 5 88基因 ) ,对 30例子宫内膜癌患者的癌组织及同一患者的正常子宫内膜组织进行分析。由两种组织中分别提取总RNA后 ,制备探针 ,与cDNA微阵列膜进行杂交 ,杂交结果经计算机软件分析 ,采用免疫组织化学的方法对结果进行鉴定。结果 (1)与细胞死亡和增殖有关的基因———IRP、Wnt 5A及癌胚抗原前体在癌组织中表达的明显增高 ;(2 )与细胞粘附、浸润有关的基因———聚集蛋白聚糖核蛋白前体在癌组织中的表达明显增高 ;(3)与浸润有关的调节因子基因———CD147、TIMP 2基因在癌组织中的表达异常增高。结论 用cDNA微阵列可同时检测两种不同组织中多个基因的差异表达 ;CD147可能与子宫内膜癌的发生与浸润有关。
Objective To investigate the mechanism of endometrial carcinoma by studying the changes of gene expression in normal endometrium and endometrial carcinoma. Methods The cDNA microarray technique (containing 5 88 genes) was used to analyze the cancer tissue of 30 patients with endometrial carcinoma and the normal endometrium of the same patient. The total RNA was extracted from the two tissues respectively. The probes were prepared and hybridized with cDNA microarray. The hybridization results were analyzed by computer software and the results were identified by immunohistochemistry. Results (1) The expression of genes related to cell death and proliferation, such as IRP, Wnt 5A and carcinoembryonic antigen precursor, were significantly increased in cancer tissues. (2) The genes related to cell adhesion and invasion were aggregated (3) The expressions of CD147 and TIMP2 genes, which are regulatory factors related to infiltration, were abnormally increased in cancer tissues. Conclusion The cDNA microarray can simultaneously detect the differential expression of multiple genes in two different tissues. CD147 may be related to the occurrence and infiltration of endometrial carcinoma.
