目的分析人直肠癌组织中转录激活因子5(ATF5)的表达变化及其与直肠癌临床病理特征的关系。方法选取四川大学华西医院胃肠外科中心2009年3~10月期间手术切除的直肠癌及距离肿瘤5 cm以远的正常直肠组织标本92对,分别应用实时荧光定量RT-PCR和免疫组织化学染色SP法检测ATF5 mRNA和蛋白的表达情况。结果直肠癌组织中33例(35.9%)ATF5 mRNA表达上调,但ATF5 mRNA相对表达量与正常直肠组织相比,二者间差异无统计学意义(P=0.363),并且ATF5 mRNA表达与患者年龄、性别、肿瘤类型、分化程度、浸润深度、淋巴结转移、远处转移及TNM分期均无关(P>0.05);但直肠癌组织中ATF5蛋白的表达高于正常直肠组织(P=0.000),并且其表达与肿瘤分化程度有关(P=0.013),而与其他临床病理因素无关(P>0.05)。结论 ATF5蛋白的表达与直肠癌的发生和分化相关,但这种相关需要进一步的研究证明。
Objective To analyze the expression of transcriptional activator 5 (ATF5) in human rectal cancer and its relationship with the clinicopathological characteristics of rectal cancer. Methods Forty-two pairs of rectal cancers resected from March to October 2009 in West China Hospital of Sichuan University and 92 normal rectal tissues distant from the tumor were detected by real-time fluorescence quantitative RT-PCR and immunohistochemical staining SP method to detect ATF5 mRNA and protein expression. Results The mRNA expression of ATF5 in 33 cases (35.9%) of rectal cancer tissues was up-regulated. However, the relative expression of ATF5 mRNA in rectal cancer tissues was not significantly different from that in normal rectal tissues (P = 0.363) (P> 0.05). However, the expression of ATF5 protein in rectal cancer tissues was higher than that in normal rectal tissues (P = 0.000), and the expression of ATF5 protein in rectal cancer tissues was significantly higher than that in normal rectal cancer tissues The expression was related to the degree of tumor differentiation (P = 0.013), but not to other clinicopathological factors (P> 0.05). Conclusion The expression of ATF5 protein is associated with the occurrence and differentiation of rectal cancer, but this correlation needs further study to prove.
