目的:探讨自体树突状细胞(DC)体外对结肠腺癌患者肿瘤引流区淋巴结(TDLN)细胞抗肿瘤活性的增强作用.方法:分离肿瘤患者的外周血单个核细胞 (PBMC)经IL-4,GM-CSF和TNF-α诱导其成熟,并以自体肿瘤冻融抗原预致敏,诱导其中的DC.另外,分离结肠癌患者的TDLN细胞, 在含有IL-2的培养体系中培养,并将TDLN 细胞分为3组:第1组为肿瘤抗原致敏的DC加 TDLN细胞(DC组);第2组冻融的肿瘤抗原加 TDLN细胞(Ag组);第3组不加DC及肿瘤抗原作为对照组(L组).比较3组TDLN细胞对结肠腺癌细胞系LS174T和黑色素瘤细胞系A375的杀伤作用.结果:DC组对LS174T细胞系的杀伤作用明显优于Ag组与L组(效靶比为20:1时,56.13%±7.33% vs 42.46%±7.68%,33.50%±7.00%, P<0.001:效靶比为10:1时,44.85%±6.50% vs 30.50%±9.17%,26.75%±8.88%,P<0.001); 而对A375细胞,各组细胞的杀伤作用没有明显差异.结论:自体DC可明显增强结肠癌患者TDLN 细胞的杀伤作用.
Objective: To investigate the antitumor activity of autologous dendritic cells (DCs) on the tumor-draining lymph node (TDLN) cells in patients with colon adenocarcinoma in vitro. Methods: Peripheral blood mononuclear cells (PBMCs) from patients with tumor were induced to mature by IL-4, GM-CSF and TNF-α and pre-sensitized with autologous tumor freeze-thaw antigen to induce DC. In addition, TDLN cells from colon cancer patients were isolated and cultured in a culture system containing IL-2, and TDLN cells were divided into three groups: DC plus TDLN cells (DC group) in which group 1 was tumor antigen-sensitized; Group of frozen-thawed tumor antigen plus TDLN cells (Ag group); Group 3 without DC and tumor antigen as a control group (L group). The cytotoxicity of TDLN cells on colon adenocarcinoma cell line LS174T and melanoma cell line A375 were compared. Results: The killing effect of DC on LS174T cell line was significantly better than that of Ag group and L group (56.13% ± 7.33% vs 42.46% ± 7.68%, 33 .50% ± 7.00%, P <0.001: 44.85% ± 6.50% vs 30.50% ± 9.17%, 26.75% ± 8 .88%, P <0.001). However, there was no significant difference in cytotoxicity between A375 cells and other groups. Conclusion: Autologous DC can significantly enhance the killing effect of TDLN cells in patients with colon cancer.
