以铈代铅作捕获剂、经实质灌注固定、液氮冷冻组织来研究小鼠肝脏葡萄糖-6-磷酸酶(G-6-Pase)。电镜观察见酶反应沉淀物位于内质网的膜上、池内及核膜上,颗粒细微、均匀,电子密度较高,未见非特异性反应产物和酶反应物弥散。反应物积聚量与细胞内酶活性相一致。细胞超微结构保存完好。
The hepatic glucose-6-phosphatase (G-6-Pase) was studied by plumbum as a capture agent after solid perfusion fixation and liquid nitrogen freezing. Electron microscopy showed that the enzyme reaction precipitates were located on the membrane of the endoplasmic reticulum, and the pellet and the nuclear membrane had fine and uniform particles with high electron density. No non-specific reaction products and enzyme reactants were not observed. Reactant accumulation is consistent with intracellular enzyme activity. Cell ultrastructure preserved.
