目的考察水飞蓟宾脂肪乳在小鼠体内的组织分布并进行肝靶向性评价。方法采用高压匀质技术制备水飞蓟宾脂肪乳,于小鼠尾静脉给药,RP-HPLC法测定各器官中不同时间的药物含量。结果与注射剂相比,水飞蓟宾脂肪乳在小鼠肝脏中的药物分布明显提高,肝的靶向指数为注射剂的21.54倍,相对总靶向效率为242.70。结论脂肪乳改变了水飞蓟宾在小鼠体内的分布,具有明显的肝靶向性。
Objective To investigate the tissue distribution of silybin-based fat emulsion in mice and evaluate its liver targeting. Methods High-pressure homogenization technique was used to prepare silybin-based fat emulsion and administered to tail vein of mice. The content of drugs in different organs at different time was determined by RP-HPLC. Results Compared with the injection, silybin oil emulsion significantly increased the distribution of the drug in the liver of the mice. The liver targeting index was 21.54 times that of the injection and the relative total targeting efficiency was 242.70. Conclusion Lipid changes the distribution of silybin in mice and has obvious liver targeting.
