应用银染PCR-SSCP方法和免疫组织化学ABC法,检测30例横纹肌肉瘤(RMS)中p53基因突变及其蛋白表达。结果显示:30例RMS中,7例SSCP阳性(23.3%)。表明这些病例相应DNA片段中发生了点突变,其中第5、6、7、8外显子SSCP阳性出现的例次分别为0、0、5、4次。p53蛋白表达率66.7%(二者符合率56.7%)。p53基因突变及蛋白表达与年龄、性别、肿瘤组织类型无关,而与肿瘤分化、转移和预后有关。提示,p53基因突变和蛋白表达可能是RMS患者预后差的因素之一,可作为判断RMS的恶性程度和预后的重要标准之一。
The p53 gene mutation and protein expression in 30 cases of rhabdomyosarcoma (RMS) were detected by silver staining PCR-SSCP method and immunohistochemistry ABC method. The results showed that in 30 cases of RMS, 7 cases were positive for SSCP (23.3%). The point mutations were found in the corresponding DNA fragments of these cases, and the occurrence of SSCP in exon 5, 6, 7, and 8 was 0, 0, 5, and 4 times, respectively. The expression rate of p53 protein was 66.7% (the coincidence rate was 56.7%). The p53 gene mutation and protein expression have nothing to do with age, gender, tumor tissue type, but related to tumor differentiation, metastasis and prognosis. It is suggested that p53 gene mutation and protein expression may be one of the factors of poor prognosis in RMS patients, and it can be used as one of the important criteria for judging the malignant degree and prognosis of RMS.
