目的比较麻黄汤等三个中医经典方剂超微饮片与传统饮片水煎液对斑马鱼胚胎毒性的影响,为其超微饮片的临床安全应用提供科学依据。方法采用模式生物斑马鱼发育正常的胚胎,在不同药物浓度药物环境中培养72 h后,于倒置显微镜下观察胚胎的存活、发育、致畸情况,72 h后,计算致斑马鱼胚胎半数死亡浓度(LC50)。结果麻黄汤超微饮片组与传统饮片组LC50分别为4.641,7.569 mg/mL;葛根芩连汤超微饮片组与传统饮片组的LC50分别为1.473,3.570 mg/mL;小柴胡汤超微饮片组与传统饮片组的LC50分别为2.223,2.441mg/mL。结论斑马鱼胚胎毒性分析结果表明,相同药物和相同浓度时,超微中药饮片毒性大于传统中药饮片。
Objective To compare the effects of three traditional Chinese medicine prescriptions such as Mahuang Decoction on the embryotoxicity of zebrafish embryos by using ultra-micro-decoction of traditional Chinese medicine decoction and traditional decoction decoction, and provide a scientific basis for its clinical application. Methods The normal embryos of model zebrafish were cultured in different drug concentrations for 72 h. The survival, development and teratogenicity of the embryos were observed under an inverted microscope. After 72 h, the median lethal concentration of zebrafish embryos (LC50). Results The LC50 values of Mahuang decoction group and traditional decoction group were 4.641 and 7.569 mg / mL, respectively. The LC50 values of Gegen Qinlian decoction group and traditional decoction group were 1.473 and 3.570 mg / mL, respectively. The LC50 of the traditional slice group was 2.223,2.441mg / mL respectively. Conclusion The zebrafish embryo toxicity analysis results show that the same drug and the same concentration, ultramicro Chinese medicine Pieces more toxic than the traditional Chinese medicine Pieces.
