为探索稻曲球不同储存时期的稻曲病菌分离最佳方法,采用新鲜稻曲球和储存7个月的稻曲球为材料,分别采取厚垣孢子悬液法和组织块分离法进行稻曲病菌的分离.结果表明:新鲜稻曲球厚垣孢子悬液法分离成功率最高达95.1%,且黄色厚垣孢子分离成功率高于墨绿色的老熟厚垣孢子;组织块分离法分离成功率最高为9.7%.储存7个月的稻曲球采用厚垣孢子悬液法分离成功率为0,改进后的组织块分离法分离成功率最高达55.1%,升汞消毒时间40 s为最佳.“,”To optimize isolation methods for Ustilaginoidea virens in different storage periods,pathogens of Ustilaginoidea virens were isolated by chlamycladosporal suspension and body tissue.The results showed that isolation rate of single spore from fresh yellow chiamydospore reached 95.1% with chlamycladosporal suspension,and it was higher than that of atroviren.But isolation rate of single spore from body tissue only reached 9.7%.The method of chlamycladosporal suspension for balls which had been stored for 7 months was defeated.55.1% was reached successfully with the modified methods and the best disinfection time with mercury chloride was 40 s.
