五步蛇蛇毒经DEAE—sephadex A—50柱层析.获得15个蛋白组分。sephadex G—50柱再层析组分Ⅱ,呈单一个蛋白峰,聚丙酰胺凝胶电泳结果呈单一区带。狗血浆纤维蛋白平板法证实五步蛇蛇毒组分Ⅱ有溶解纤维蛋白的作用,同时狗血浆纤维蛋白热平板法还证实其溶解纤维蛋白的作用系直接性的。体外试管实验显示组分Ⅱ虽对纤维蛋白和纤维蛋白原均有溶解作用.但在浓度17.8mg·L~(-1)以下时.其溶解纤维蛋白活性相对较高。兔scO.1ml(500mg·L~(-1)组分Ⅱ未发现类似五步蛇全毒的出血反应。小白鼠腹腔注射纯化后的组分Ⅱ,LD_(50)为11.105 mg,kg~(-1).
Five-step snake venom by DEAE-sephadex A-50 column chromatography to obtain 15 protein components. Dog plasma fibrin plate method confirmed that the five-step snake venom fraction II has the effect of dissolving fibrin, while the dog plasma fibrin plate method also confirmed that the role of fibrinolytic system is straightforward. In vitro tube experiments showed that although component Ⅱ had a dissolution effect on fibrin and fibrinogen, its soluble fibrin activity was relatively high at a concentration below 17.8mg · L -1. Rabbit scO.1ml (500mg · L -1) did not find a component Ⅱ similar to the whole body hemorrhagic reaction of pentobarbital mice were purified by intraperitoneal injection of fraction Ⅱ, LD_ (50) was 11.105 mg, kg ~ ( -1).
