【目的】依据形态学、生理生化和26S rDNA D1/D2区序列特征,确定一株胡杨内生酵母菌的分类学地位。【方法】通过平板划线分离法分离纯化胡杨树干部内存液样品中的内生酵母菌;检测分离菌株形态学特征和生理生化特征,进一步对酵母菌株的26S rDNA D1/D2区序列进行扩增以及系统进化分析。【结果】传统的形态学特征和生理生化特征鉴定及26S rDNA D1/D2区序列分析结果表明菌株ZA-7与Rhodotorulaminuta(FR870029)相近,相似率为99.4%,存在4个碱基的差异。【结论】从胡杨树干液中分离得到了一株尚未从植物中分离到的内生酵母菌,并将该菌株鉴定为Rhodotorula minuta的近似种。
【Objective】 The taxonomic status of an endophytic Saccharomyces cerevisiae was determined according to morphological, physiological and biochemical characteristics and 26S rDNA D1 / D2 region sequence. 【Method】 The endophytic yeasts in the samples of Populus euphratica from the trunk of Populus euphratica were isolated and purified by the plate scribing separation method. The morphological characteristics and physiological and biochemical characteristics of the isolated strains were tested. The 26S rDNA D1 / D2 sequences of the yeast strains were further expanded Increase and systematic evolutionary analysis. 【Result】 The results of traditional morphological, physiological and biochemical characteristics analysis and sequence analysis of 26S rDNA D1 / D2 showed that strain ZA-7 was similar to Rhodotorula minuta (FR870029) with a similarity of 99.4% with a 4-base difference. 【Conclusion】 An endophytic yeast that has not been isolated from the plant was isolated from Populus euphratica, and the strain was identified as an approximate species of Rhodotorula minuta.
