目的:研究Th17细胞及其相关细胞因子白细胞介素-17(IL-17)在胶原诱导性关节炎(collagen-induced ar-thritis,CIA)小鼠体内的变化。方法:利用Ⅱ型胶原蛋白建立小鼠CIA模型。应用流式细胞术(FCM)分析小鼠脾脏及引流淋巴结中Th17细胞的比例,采用酶联免疫吸附试验(ELISA)检测小鼠血清中IL-17的含量。结果:CIA小鼠脾脏及引流淋巴结中Th17细胞的比例分别为(3.19±0.70)%、(3.54±0.97)%,显著高于正常对照组的(0.95±0.34)%、(1.31±0.47)%,P<0.01。CIA小鼠血清中IL-17的含量也明显高于正常对照组(P<0.05)。结论:Th17细胞及其分泌的细胞因子IL-17在CIA小鼠体内明显升高,并与疾病的发生密切相关。
AIM: To investigate the changes of Th17 cells and its related cytokine interleukin-17 (IL-17) in collagen-induced arthritis (CIA) mice. Methods: Mouse CIA model was established by using type Ⅱ collagen. The proportion of Th17 cells in the spleen and draining lymph nodes was analyzed by flow cytometry (FCM). The level of IL-17 in sera of mice was detected by enzyme linked immunosorbent assay (ELISA). Results: The proportion of Th17 cells in spleen and draining lymph node of CIA mice was (3.19 ± 0.70)% and (3.54 ± 0.97)%, respectively, which was significantly higher than that of the normal control group (0.95 ± 0.34%, 1.31 ± 0.47% , P <0.01. The content of IL-17 in serum of CIA mice was also significantly higher than that of normal control group (P <0.05). Conclusion: Th17 cells and its secreted cytokine IL-17 in CIA mice significantly increased, and closely related to the occurrence of the disease.
