用人类大肠癌细胞株HI29作抗原,融合成三株杂交瘤细胞(Hb_3·Ab_5和Ad_5)。用间接免疫荧光检查,发现这三株细胞产生的抗体,对人类结肠癌组织反应的阳性率分别为86%、45%和58%。三株抗体也对胃癌、胰腺癌和肺癌反应阳性,但对肾癌和各种肉瘤组织阴性。三株抗体对正常结肠上皮阴性,所以能鉴别结肠癌和正常大肠上皮组织,具有相对的器管特异性。此外,抗体对A、B、O型血及人类皮下成纤
Using the human colorectal cancer cell line HI29 as an antigen, three hybridoma cells (Hb_3·Ab_5 and Ad_5) were fused. Using indirect immunofluorescence, the antibody produced by these three cells was found to be 86%, 45%, and 58% positive for human colon cancer tissues. The three antibodies were also positive for gastric cancer, pancreatic cancer, and lung cancer, but negative for kidney cancer and various sarcoma tissues. The three antibodies were negative for normal colonic epithelium, so they were able to identify colon cancer and normal large intestine epithelium with relative tube specificity.
