应用UPLC-MS-MS系统研究盐酸阿比朵尔在实验动物大鼠体内的代谢情况,利用超高效液相色谱的分离能力结合串联质谱的质量分辨性能,以药物分子结构中的同位素现象为指针,共检测和鉴定出盐酸阿比朵尔在大鼠体内的代谢产物9种,其中Ⅰ相代谢产物2种,Ⅱ相代谢产物7种,并包括两对非对映异构体。实验结果揭示药物在大鼠体内主要经过N-脱烷基化、S-氧化、O-葡糖醛酸轭合和O-硫酸酯化轭合等代谢反应。
The UPLC-MS-MS system was used to study the metabolism of abiraterone hydrochloride in rats. The separation ability of ultra-high performance liquid chromatography coupled with the mass resolution of tandem mass spectrometry was used as a guideline for the isotope phenomenon in the molecular structure of the drug A total of 9 metabolites were detected and identified in Arabidopid hydrochloride rats, including 1 phase Ⅰ metabolites and 7 phase Ⅱ metabolites, including two pairs of diastereomers. The experimental results revealed that the drug mainly undergoes metabolic reactions such as N-dealkylation, S-oxidation, O-glucuronide conjugation and O-sulfation conjugation in rats.
