目的探讨松花粉缓解砷中毒小鼠大脑皮质细胞凋亡的机制。方法雌性昆明小鼠随机分为对照组(饮用蒸馏水)、干预组(NaAsO2:5μg/g;松花粉:1 g/kg)、NaAsO2组(NaAsO2:5μg/g),每组8只。每天1次,连续45 d,运用ELISA方法和Western Blot技术检测小鼠大脑皮质细胞凋亡相关因子BAX、Fas、Bcl-2和P38/MAPK14蛋白的表达量。结果 NaAsO2组BAX、Bcl-2含量均高于对照组和干预组,差异有统计学意义(P<0.05);NaAsO2组Fas含量和P38/MAPK14蛋白的表达量高对于对照组,差异有统计学意义(P<0.05)。结论松花粉降低NaAsO2中毒小鼠大脑皮质细胞凋亡主要是通过下调BAX和通过Fas途径的Bcl-2因子。
Objective To investigate the mechanism of pine pollen ameliorating the cerebral cortical cell apoptosis induced by arsenic poisoning in mice. Methods Female Kunming mice were randomly divided into control group (drinking distilled water), intervention group (NaAsO2: 5μg / g; pine pollen: 1g / kg) and NaAsO2 group (NaAsO2: 5μg / g) Once a day for 45 consecutive days. The expressions of BAX, Fas, Bcl-2 and P38 / MAPK14 in mouse cerebral cortex were detected by ELISA and Western Blot. Results The levels of BAX and Bcl-2 in NaAsO2 group were significantly higher than those in control group and intervention group (P <0.05). The levels of Fas and P38 / MAPK14 protein in NaAsO2 group were significantly higher than those in control group Significance (P <0.05). Conclusion Pine pollen can reduce the apoptosis of cerebral cortex cells induced by NaAsO2 in mice by down-regulating BAX and Bcl-2 through Fas pathway.
