目的 研究大肠杆菌表达的汉滩病毒囊膜糖蛋白G1和 G2的免疫原性和免疫保护作用 .方法 将大肠杆菌表达的 G1和 G2免疫小白鼠 ,测定免疫小鼠血清中汉滩病毒抗体的间接免疫荧光抗体和中和抗体滴度并利用被动免疫保护试验观察 G1和 G2免疫小鼠血清和脾细跑对汉滩病毒致死性感染乳鼠的保护作用 .结果 表达 G1和 G2免疫小鼠血清中抗汉滩病毒抗体的滴度分别为 1∶ 16 0 ,1∶ 32 0 ;G1,G2共同免疫不能提高免疫效果 ;NP,G1,G2共同免疫 ,血清中抗汉滩病毒抗体的滴度较 NP单独免疫低 ;有一份 G2免疫血清中检测到低效价的中和抗体 ,效价为 1∶ 10 ;G1,G2免疫血清及免疫小鼠脾细胞对致死性感染的乳鼠都有一定保护作用 ,且 G2较明显 .结论 表达的 G2能刺激小白鼠产生中和抗体 ,但表达的 G1不能刺激产生可测定的中和抗体 ,表达的G1,G2和 NP共同免疫不能提高中和抗体的滴度 ;G1和 G2免疫血清和脾细胞都能被动保护致死性感染的乳鼠 ,或使乳鼠免于死亡或延长其存活时间 ,G2的保护作用更明显 ,G2免疫血清的保护率可达 5 0 % ,脾细胞的保护率可达 10 % .
Objective To study the immunogenicity and immunoprotection of Hantaan virus envelope glycoprotein G1 and G2 expressed in Escherichia coli.Methods G1 and G2 cells expressed in E. coli were immunized with mouse and the indirect antibody of Hantaan virus Immunofluorescent antibody and neutralizing antibody titers were used to observe the protective effect of G1 and G2 immunized mice serum and splenic lymphocytes on Hantavirus virulence infected neonatal rats by passive immunoprotection assay.Results The expression of G1 and G2 immunized mice serum Anti-Hantaan antibody titers were 1:16 0, 1: 32 0; G1, G2 co-immunization can not improve the immune effect; NP, G1, G2 co-immunization, serum anti-Hantaan virus titer than the NP Immunized with low titer of neutralizing antibody in G2 immune serum with a titer of 1:10; G1 and G2 immune sera and splenocytes from immunized mice were protective against lethally infected neonates , And G2 was more obvious.Conclusion G2 can stimulate neutralizing antibodies in mice, but the expression of G1 can not stimulate the production of measurable neutralizing antibodies, and the co-immunization of G1, G2 and NP can not increase the amount of neutralizing antibodies ; G1 and G2 immune sera and spleen cells can passively protect the deadly infected suckling mice or prevent or prolong the survival time of suckling mice, the protective effect of G2 is more obvious, and the protective rate of G2 immune sera can reach 50 %, Spleen cell protection rate of up to 10%.
