目的在观察仙茅对虚寒大鼠神经内分泌功能改善基础上,探讨辛热药仙茅对虚寒状态动物肝脏药物代谢酶的影响及在辛热药性表达中的意义。方法将SD大鼠随机分为空白对照组,虚寒模型组,仙茅大、中、小剂量组。以氢化可的松(3.75 mg/kg)肌肉注射复制虚寒大鼠模型,辛热药仙茅以30、20、10 g/kg干预,红霉素-N脱甲基法检测动物肝脏代谢酶细胞色素P4503A(CYP3A)活性,放免法检测动物血浆环磷酸腺苷(cAMP)、环磷酸鸟苷(cGMP)含量,Western blot法检测动物肝脏孕烷X受体(PXR)蛋白表达,免疫组化法检测动物肝脏蛋白激酶A(PKA)蛋白表达。结果虚寒大鼠肝脏药物代谢酶活性降低,PXR表达减少,cAMP、cGMP水平降低,PKA表达减少,仙茅各剂量组可提高CYP3A活性和cAMP含量,增强PXR、PKA表达,对cGMP影响不大。结论仙茅可能通过调节cAMP-PKA信号通路改善虚寒状态的代谢功能低下症状,从而表明辛热药的部分药性实质。
OBJECTIVE To observe the effect of Curculigo on neuroendocrine function of asthenic rats and to explore the effect of Curculigo orchidensis on hepatic drug metabolizing enzymes in asthenic animals and its significance in the expression of Herb. Methods SD rats were randomly divided into blank control group, Deficiency model group, Curculigo, middle and small dose group. The model of asthenia cold was induced by intramuscular injection of hydrocortisone (3.75 mg / kg). The effects of 30, 20 and 10 g / Cytochrome P4503A (CYP3A) activity was assayed by radioimmunoassay. The plasma levels of cAMP and cGMP were determined by radioimmunoassay. The expression of PXR protein was detected by Western blot. Immunohistochemistry Method was used to detect the protein expression of protein kinase A (PKA) in liver of animals. Results Compared with the control group, the hepatic metabolism enzyme activity, the decrease of PXR, the decrease of cAMP and cGMP, and the decrease of PKA expression were found in each group. Curculigo oryzae could increase the activity of CYP3A and cAMP, enhance the expression of PXR and PKA, and had little effect on cGMP . CONCLUSION Curculigo may improve the hypometabolic symptoms of asthenopia by regulating the cAMP-PKA signaling pathway, indicating some of the medicinal properties of the medicinal herbs.
